I shot this picture in AV mode, it was overcast so i had set the appropriate white balance and selected a wide aperture so the background is out of focus. The only thing is that it didn't seem to be making much of a difference when i selected ISO as 160 or 600! Why is that? How can you know what ISO will be your best choice for the circumstances?

The lighting works well, with the light evenly distributed, so that the portrait has come out in lighter tones, the gentle shadows outlining the form of the young couple.
The shutter speed is fast enough that none of that sharpness has been lost.

There is no need to raise the ISO, this will only bring about an increase in noise within the frame, and the shutter speed then has to be faster. Because the portrait is static, you don’t need to use a very fast shutter speed, meaning the ISO can be kept to a minimum. Had the people been moving within the frame, you would have needed to raise the ISO a little and increase the shutter speed to retain the sharpness.
You really want to have the faces fully lit, to show that glint in the eye, and bring that gaze to life. To do this, you could use a reflector or add flash, lowering the intensity beforehand in the camera menu.
